AN EFFECT OF DIETARY PROTEIN ON THE TOXICITY OF EDECRIN(ETHACRYNIC ACID) IN RATS

Abstract

|
The effect of ethacrynic acid treatment on the mortality, liver weight, glutathione content and activity of its conjugating enzyme, glutathione S-transferase, in liver of rats previously fed low or standard protein diet, was studied.
The ratio of liver weight per body weight in rats fed low protein, showed an increasing tendency, but that of rats fed standard protein diet showed no difference from the control group.
After injection of ethacrynic acid to the rats fed low protein diet, the mortality was higher than those fed standard protein diet. The content of glutathione and activity of glutathione S-transferase in liver of rats fed standard protein diet were determined as higher, compared with those fed low protein diet, but an increasing rate of liver glutathions S-transferase activity in rats fed low protein diet was more or less increased by the injection of ethacrynic acid than those fed standard protein diet.
These results indicate that a side effect of ethacrynic acid on the rat liver may have relevance to the dietary protein.
